Production of Saccharomyces boulardii biomass using sesame cake: A preliminary investigation

Saccharomyces boulardii is a variety of Saccharomyces cerevisiae that is usually indicated as Saccharomyces cerevisiae var.boulardii. S. boulardii is non-pathogenic and vegetative yeast, which is extensively used as probiotic. Several studies showed that S. boulardii has vital therapeutic role in the host such as prevention of antimicrobial associated diarrhoea. Therefore, there were huge numbers of studies that have focused on optimization of biomass production of S. boulardii under different conditions (defined and semi-defined media, dilution rate, nitrogen content, substrate level, glucose content, dissolved oxygen content, pH, etc). Different by-products that come from the food industries have been investigated as nutrient source for production of biomass of probiotics. Sesame cake is considered is by-product, derived from sesame seed pressing to extract oil. Palestinian sesame, derived-food-industry, has a significant market. Until now, sesame cake as by products is not well exploited. The aim of this study is to evaluate the possibility to use sesame cake as nutrient source for S. boulardii. As a matter of fact, the strain Saccharomyces boulardii CNCM I-1079 was inoculated in a broth constituted by a water solution at different concentrations of sesame cake (1, 2, 3 and 5% w/v). Growth was evaluated by viable counting on Potato Dextrose Agar (PDA) at different time (i.e. 0,1,2,4,5,6, 8, 24 h). In the same way, the growth of S. boulardii was evaluated on potato dextrose broth as standard medium for comparison purposes. Our preliminary findings showed that the growth of S. boulardii at 5% of sesame cake was similar to that performed in the standard medium. In particular, growth curve in both medium showed a lag phase of about 2 h and a log phase of about 5 h until a maximum population level of about 5x108 CFU/ml was reached. In conclusion, the finding of the study showed that sesame cake is a promising by-product as feed source for growth of S. boulardii without further modifications.
