EFFECT OF SOME ANTIBIOTICS ON THE ACTIVITY OF GLUTATHIONE-S-TRANSFERASES: AN IN VITRO STUDY

Introduction

Serious bacterial infections can be life threatening and require prompt treatment with antibacterial agents like antibiotics. However, the use of antibiotics may also generate adverse effects in patients including inhibition of the activity of some enzymes like Glutathione-S-transferases (GSTs). These isoenzymes are widely distributed in nature and present in different organisms such as microbes, plant, fish, insect, birds and mammals. Also, they play an important role in the metabolism and detoxification of numerous endobiotics and xenobiotics. Based on the importance of these isoenzymes they have been extensively investigated in mammals. So, the following experiment was performed to explore the effect of five antibiotics on GSTs activity.

 

Methods

Glutathione-S-transferases were extracted and purified from sheep liver using ammonium sulfate precipitation, gel filtration and affinity column chromatography. The effect of Amikacin, Aztreonam, Cephalothin, Clindamycin and Doxycycline on GSTs activity was studied by a spectrophotometric method using 1-chloro-2,4-dinitro-benzene (CDNB) as substrate.

 

Results

The obtained results revealed that all antibiotics under study demonstrated inhibitory effect on GSTs activity in the tested concentrations range (0.1-10 mM). Moreover, doxycycline found to display an inhibitory effect on the activity of GST at low concentration )0.1, 0.25, 0.5, 1 and 2.5 mM), which resulted in 32.8, 44,50.7  and 100 % inhibition respectively. Results also provided that the doxycycline concentration required to reduce the GSTs activity to half is 0.135 mM which is lower than the other screened antibiotics.

 

Conclusions

Antibiotics are potential inhibitors for metabolic enzymes. Therefore, it is necessary to determine the doses of these antibiotics for microbial treatment with no adverse effect on the activity of essential enzymes.
